Résumé: For many industrial applications, the combination of two different monomers in statistical or diblock copolymers enhances the properties of the corresponding polymer. However, during the polymerization reaction, homopolymers might be formed and can influence the properties for the applications. Consequently, the separation and the quantification of the homopolymers contained in copolymer samples are crucial. In addition, the charge density distribution of the statistical copolymer is an important characteristic for the applications. The purpose of this work was to study the characterization of a statistical copolymer of acrylic acid (AA) and diallyldimethyl ammonium chloride (DADMAC) by capillary electrophoresis (CE) in acidic conditions (cationic copolymers). For that purpose, a free solution electrophoretic separation was carried out according to the charge rate (chemical composition) independently of the molar mass. The second objective was to compare contactless conductivity detection and indirect UV absorbance modes for the quantification of DADMAC homopolymers present in copolymer samples. Different coated capillaries based on neutral or positively charged modification were also compared. The comparison of indirect absorbance UV and contactless conductimetric detection demonstrated that both detection modes can be used for a complete CE characterization of non-UV absorbing PAA-DADMAC copolymers.

Résumé: The four first generations of dendrigraft poly-L-lysine have been studied in dimethylformamide (aprotic solvent) and in 0.2 M NaCl aqueous solutions by isothermal translation diffusion, 1H NMR and viscometry methods. The relationships between diffusion coefficient, intrinsic viscosity and molar mass have been determined for dendrigraft poly-L-lysines, and the scaling index values have been compared to classical trifunctional dendrimers. Dendrimers and dendrigraft poly-L-lysines exhibited similitudes in their hydrodynamic behaviors. Nevertheless, dendrigraft poly-L-lysines displayed a specific behavior in solution. In contrast to dendrimers, a significant change of hydrodynamic dimension of dendrigraft poly-L-lysines according to the nature of the solvent has been observed. In aprotic solvent, the dendrigraft poly-L-lysine dimensions are about two times lower than in aqueous media (i.e., the hydrodynamic volume is contracted by a factor 8 in dimethylformamide), revealing the softness of dendrigraft poly-L-lysine compared to classical trifunctional dendrimers.

Résumé: The "coarse-grained" bead modeling methodology, BMM, is generalized to treat electrostatics at the level of the nonlinear Poisson-Boltzmann equation. This improvement makes it more applicable to the important class of highly charged macroions and highly charged peptides in particular. In the present study, the new nonlinear Poisson-Boltzmann, NLPB-BMM procedure is applied to the free solution electrophoretic mobility of low molecular mass oligolysines (degree of polymerization 1-8) in lithium phosphate buffer at pH 2.5. The ionic strength is varied from 0.01 to 0.10 M) and the temperature is varied from 25 to 50°C. In order to obtain quantitative agreement between modeling and experiment, a small amount of specific phosphate binding must be included in modeling. This binding is predicted to increase with increasing temperature and ionic strength.

Résumé: Our goal was to design carbohydrate mimetics capable of preventing cell adhesion of pathogens by targeting lectins. The original synthesis of these mimetics combines the automated chemistry of DNA and ''click'' chemistry. From simple building blocks (i.e. phosphoramidites, solid supports and carbohydrates such as alkyne or azide derivatives), a ''Lego'' approach provides complex and varied decoys exhibiting various topologies and number of carbohydrate residues. The resulting glycomimetics tagged with a specific DNA sequence are efficiently immobilized on a DNA microarray by double strand formation. This glycoarray allows the study of the interactions between carbohydrate mimics and lectins using a minute amount of material. This micro system using DNA arrays is much more effective than conventional carbohydrate microarrays. The studies allowed the identification of the important structural parameters for a customized construction of high-affinity carbohydrate mimics.

Résumé: We have evaluated the laser desorption ionization mass spectrometry (LDI-MS) performance of six nanostructured silicon surfaces of different morphologies and chemical functionalizations. The substrates have been synthesized either by metal-assisted etching method or by vapor−liquid−solid (VLS) growth technique. In addition to the commercial nanostructured silicon-based surface (NALDI) target plates, serving as reference, the homemade surfaces have been evaluated in mass spectrometry experiments conducted with peptide solutions mimicking tryptic digests. LDI surfaces synthesized by metal-assisted etching method were the most efficient in terms of signal intensities and number of detected peptides. The surface providing the best LDI-MS performance was composed of two nanostructured layers. Interestingly, we also observed a significant influence of the type of organic coating (hydrocarbon vs fluorocarbon) on peptide ionization discrimination.

Résumé: The present study was conducted to assess the structures of the main unknown oxygenated metabolites of EAPB0203. The first step was to assign all the 1H and 13C NMR of both EAPB0203 and its demethylated metabolite (EAPB0202) to the corresponding atoms in their molecular structures and to elucidate the fragmentation pathways for the [M + H] + ions of these compounds using high-resolution mass spectrometry (MS). MS/MS spectra showed that both protonated molecules possessing an even number of electrons were unexpectedly losing radicals such as H*,CH3*, or even C7H7* giving stable radical cations. In vitro metabolism studies were investigated in rat and dog liver microsomes and in the filamentous fungus Cunninghamella elegans. Structural elucidation of six oxygenated metabolites was performed based on the following: (i) their fragmentation pathways in liquid chromatography−MS/MS (LC-MS/MS) analyses; (ii) comparison of their changes in their molecular masses and fragment ions with those of the parent drugs; and (iii) the results of online H/D exchange experiments that provided additional evidence in differentiating hydoxylated metabolites from N-oxides. Structures of the metabolites were elucidated by LC-MS/MS and comparison with synthetic standards; structures of these standards were confirmed using one- and two-dimensional 1H NMR spectroscopies.

Résumé: After extra-cellular stimulation of G-Protein Coupled Receptors (GPCRs), GDP/GTP exchange appears as the key, rate limiting step of the intracellular activation cycle of heterotrimeric G-proteins. Despite the availability of a large number of X-ray structures, the mechanism of GDP release out of heterotrimeric G-proteins still remains unknown at the molecular level. Starting from the available X-ray structure, extensive unconstrained/constrained molecular dynamics simulations were performed on the complete membrane-anchored Gi heterotrimer complexed to GDP, for a total simulation time overcoming 500 ns. By combining Targeted Molecular Dynamics (TMD) and free energy profiles reconstruction by umbrella sampling, our data suggest that the release of GDP was much more favored on its phosphate side. Interestingly, upon the forced extraction of GDP on this side, the whole protein encountered large, collective motions in perfect agreement with those we described previously including a domain to domain motion between the two ras-like and helical sub-domains of Ga.
