Solvent-free Peptide Synthesis

Chemists from the Department of Aminoacids, Peptides and Proteins of the IBMM have developed the first solvent-free method for making a peptide bond by employing a ball-mill grinder. In the absence of solvent, the ball-mill was used to couple urethane protected aminoacid N-carboxyanhydride and an aminoester to produce several dipeptides and a tripeptide. Sweetener aspartame was synthesized using this method. This represents a greener approach to the preparation of peptides, particularly as these compounds become more popular as pharmaceuticals.
