Modified oligonucleotides
In addition to the implication of the teachers-researchers of the team in various teachings in the University of Montpellier II and in Sète IUT, several members of the team, researchers as well as teacher-researchers are involved in the “Oligonucleotides and Analogues” module for the Master 2 on “Biomolecular Engineering” Research, a course also given in the 3rd year of the Ecole Nationale Supérieure de Chimie of Montpellier.
Programmes of the various teachings : The objective of these teachings is to lay the bases of oligonucleotides chemistry, of their analogues as well as to provide an insight into their biological applications. The protective moieties of the different functions of nucleotides as well as the different formation methods for internucleosidic bonds are examined. Syntheses, either solution convergent or linear on solid support, are developed. The main purification and characterisation techniques of these compounds are examined. Finally, the synthesis and the use of oligonucleotides linked covalently to markers and effectors are illustrated.
1) Biological motivations
2) Background of chemistry of nucleic acids
3) Protective moieties in DNA synthesis
4) Protective moieties in RNA synthesis
5) Formation of the internucleosidic bond
6) automated supported synthesis
7) Purification and characterisation of oligonucleotides
8) Oligonucleotide analogues
9) Attachment of markers and effectors
